Human Technopole (HT) is an interdisciplinary life science research institute, created and supported by the Italian Government, with the aim of developing innovative strategies to improve human health. HT is composed of five Centers: Neurogenomics, Computational Biology, Structural Biology, Genomics, and Health Data Science. The Centers work together to enable interdisciplinary research and to create an open, collaborative environment that will help promote life science research both nationally and internationally.
The Genomics Centre at HT is dedicated to research aimed at uncovering the complex mechanisms governing gene expression and how heritable genetic information translates into phenotypic traits. Applied to humans, and in the context of precision medicine, this type of research can identify molecular targets and markers for disease prevention, early detection, and personalised treatment. In addition to carrying out genetic and genomic studies with a focus on (but not limited to) disease-associated mechanisms, the Genomics Research Centre will promote and help implement large-scale genomic screening projects for patient stratification.
We are seeking a motivated Curricular Intern to join our team in the Soranzo Group - Human Technopole for a period of minimum of 10 months.
The intern will be involved in a project whose aim is to functionally characterize the impact of germline variation in the development and progression of clonal hematopoiesis (CHIP) and related complications.
Prioritized germline variants will be engineered by CRISPR/ Cas9 technology in relevant cell models such as hES Cs (human embryonic stem cells) together with the classical CHIP somatic mutations. Cells will be genotyped, expanded and then differentiated towards the relevant hematopoietic lineages (megakaryocytes, erythrocytes, lymphocytes, etc. . ). The effect of the variant will be assessed by RNA sequencing and by tailored functional assays (i. e. apoptosis, cell cycle, proliferation, migration, colony formation, etc).
Key findings will then be validated in primary CD34+ hematopoietic cells (HS Cs).
Your main tasks and responsibilities
•
Maintenance, engineering and differentiation of stem cells;
•
Cells characterization by multiparametric flow cytometry and single cell RNA+ATAC sequencing;
•
Support omics analysis.
Essential requirements
•
Currently a Master student (biotechnology, cancer biology, cell and molecular biology or similar);
•
Previous lab experience;
•
Theoretical knowledge in genetics and cancer biology.
Preferred requirements
•
Previous experience in cell culture;
•
Basic knowledge of R studio.
Organizational and social skills
•
Excellent communication and team working skills;
•
Capable of handling multiple assignments effectively.
